Resumo: Stroke is a major cause for morbidity and mortality in developed countries. Given the increased life expectancy of populations, finding ways of preventing the disease and adequate treatment is a priority task. For this purpose, it is warranted that risk factors, genetic and environmental, are properly characterized, both for disease susceptibility and for disease outcome. The objective of the present project is the identification and characterization of genetic factors predisposing to stroke and influencing disease outcome and patient’s prognosis, assessed 3 and 12 months after a stroke episode, in a large population sample of stroke patients and ethnically matched controls. For this purpose, the study design will include the association analysis of functional candidate genes, with a particular focus on genes involved in neuroinflammation and neuronal survival and regeneration, and a high resolution whole-genome association study in pooled DNA samples. Functional analysis will be carried out to determine the normal and pathogenic function of identified genes.
